Walnut oligopeptides (WOPs) are a class of small molecules with significant biological activity extracted from walnuts (Juglans regia L.) through enzymatic hydrolysis technology. Its essence is a short chain peptide segment composed of 2 to 10 amino acids, with a molecular weight typically less than 500 daltons, coexisting in the form of free amino acids, oligopeptides, peptides, and a small amount of proteins.
Its walnut peptide contains 18 amino acids, including 8 essential amino acids, among which glutamic acid has the highest content, followed by arginine. Glutamate, as one of the fundamental amino acids in human nitrogen metabolism, is particularly important for brain tissue - brain tissue can only oxidize and utilize glutamate instead of other amino acids. Therefore, glutamate can serve as an energy source for brain tissue, providing energy for the brain.

This small molecule structure gives WOPs a core advantage: they can be directly absorbed by the human body without digestion, and their absorption efficiency is much higher than that of single amino acids or intact proteins. This characteristic has attracted much attention in the fields of functional foods and nutritional health.

Walnut oligopeptides, the biologically active essence extracted from the "brain nourishing fruit" of walnuts, are becoming one of the most promising star raw materials in functional food and general health industry with its six core effects of neuroprotection, cardiovascular protection, immune regulation, intestinal maintenance, skin anti-aging, anti fatigue and anti-oxidation.

1. Brain health and neuroprotection - the most dazzling star effect
The performance of walnut oligopeptides in the field of brain health is truly stunning.
(1) Improve learning and memory abilities
An experiment on juvenile mice (48 male C57BL/6J mice) showed that the mice were randomly divided into a blank control group and three walnut peptide intervention groups: low (110mg/kg · bw), medium (220mg/kg · bw), and high (440mg/kg · bw).
After continuous intervention for 30 days, Morris water maze, jumping platform experiment, and shuttle box experiment were conducted. The results show:
Morris water maze: The escape latency of the high-dose group was significantly shortened compared to the control group (P<0.05), and the residence time and number of passes in the target quadrant were significantly increased in the medium and high-dose groups (P<0.05);

Jumping Platform Experiment: The low and high dose groups showed a decrease in the number of errors during the 24-hour test (P<0.05), while all three intervention groups had an increase in latency (P<0.05). The medium dose group had a slower memory loss of electric shocks (P<0.05);
Shuttle box experiment: The low, medium, and high dose groups showed an increase in the number of active avoidance attempts (P<0.05) and a decrease in the number of electric shocks received (P<0.05).
The conclusion is clear: it can significantly enhance the learning and spatial memory ability, active and passive avoidance ability, and may delay the decline of learning and memory in young mice.
The mechanism is closely related to antioxidant activity - the walnut peptide dose group has higher serum T-SOD and GSH Px activities than the control group, and significantly lower MDA levels than the control group (P<0.05).
In addition, a study published in Food&Function successfully isolated a novel oligopeptide AQ from the hydrolysate of defatted walnut meal, and identified its chemical structure as AASDCQ. This peptide exhibits excellent free radical scavenging ability at a concentration of 1mg/mL: O ₂⁻ free radical scavenging rate of 81.00%, DPPH free radical scavenging rate of 79.40%, and ABTS free radical scavenging rate of 67.09%.

More importantly, AQ has a strong protective effect against H-SY5Y neuronal damage induced by H ₂ O ₂, significantly reducing cell damage and apoptosis. In vivo experiments have also shown that AQ can promote angiogenesis in quail chorioallantoic membrane and reduce ROS accumulation and prolong lifespan in the C. elegans model.
Another clinical study has shown that it can improve age-related learning and memory disorders, and is considered a potential and effective intervention.

(2) Deep mechanisms of neuroprotection
AQ oligopeptides also exhibit potent anti-inflammatory and neuroprotective effects: in LPS stimulated RAW264.7 cells, AQ effectively inhibits the secretion of pro-inflammatory factor NO (IC ₅₀=46.03 ± 0.32 μ M) and suppresses the expression of IL-6 and TNF - α. Western Blot further confirmed its anti-inflammatory mechanism. Overall, AQ possesses four layers of neuroprotective properties: antioxidant, anti-inflammatory, pro angiogenic, and anti-aging.

2. Cardiovascular health - the natural shield that protects the heart
The protective effect of walnut oligopeptides on the cardiovascular system has been validated by multiple sources. Its mechanism of action includes:
Reduce levels of "bad" cholesterol (LDL);
Regulating blood pressure;
Improve blood circulation.
These benefits stem from the synergistic effect of healthy fats, antioxidants, and essential amino acids in WOPs. In the context of increasingly severe global cardiovascular disease, this natural heart protection solution has significant public health implications.

3. Immune regulation - activating the human defense corps
Research has shown that it can significantly improve humoral immunity and cell-mediated immune responses, manifested as:
Enhancing macrophage phagocytic function: In the zebrafish model, powder has been shown to promote macrophage phagocytosis (WO2017124921A1 patent record);
Enhance the activity of natural killer cells (NK cells).
This means that it is not only a "nutritional supplement", but also an immune modulator that can activate the innate immune system of the human body in all aspects.

4. Gut Health - Probiotic like "Nourishing Bacteria" Effect
WOPs have prebiotic properties and can serve as a nutritional source for beneficial gut microbiota, promoting the growth and reproduction of "friendly microorganisms" such as bifidobacteria and lactobacilli. The study also found that WOPs can promote bacterial growth and reproduction, increase bacterial biofilm and extracellular polysaccharide (EPS) secretion. By maintaining a healthy gut microbiota ecology, it indirectly enhances digestive function and nutrient absorption efficiency.

5. Skin Health - The Anti Aging Code from Inside Out
In the field of cosmetics, walnut oligopeptides also shine brightly:
Stimulating the proliferation of keratinocytes: These cells are responsible for producing keratin, the core component of the skin's protective barrier, which promotes skin repair and regeneration;
Promote the synthesis of collagen and elastin: These two proteins are key to maintaining skin elasticity and firmness, and can effectively combat skin sagging and aging;
Dual protection of antioxidant and anti-inflammatory: reducing the damage of free radicals to the skin and delaying chronic aging.

6. Anti fatigue and comprehensive antioxidant
It has also been proven to have anti fatigue ability and strong antioxidant capacity. Researchers have found that it can eliminate hydroxyl radicals and superoxide anion radicals, which is the molecular basis for its various health benefits.

The preparation of WOPs mainly adopts multi-step processes such as enzymolysis - ultrafiltration - gel filtration chromatography. Specifically, the defatted walnut meal was first used as raw material, enzymatically hydrolyzed by protease, then the components with different molecular weights were separated by ultrafiltration membrane, and finally the target oligopeptides were purified by gel filtration chromatography.
Taking the preparation method of the test solution described in patent WO2017124921A1 as an example: Accurately weigh 10mg of WOP powder, place it in a 50mL brown volumetric flask, add 30mL of ethanol, sonicate for 5 minutes, dilute with ethanol to the mark, and shake well. This indicates that WOP powder has good ethanol solubility.
In terms of product form, the market is mainly divided into four types: liquid type, dry powder type, peptide type, and oligopeptide type. Liquid type has excellent solubility and is suitable for beverages and health supplements, with rapid absorption; Dry powder type (i.e. walnut oligopeptide powder) is easy to store and transport, suitable for food formulas and dietary supplements. It appears as a fine white to milky yellow powder, without clumping or impurities, with a purity of over 95% or even 99%.
